Washington, November 2 (ANI): An injection of banked sperm-producing stem cells can restore fertility in male primates who become sterile due to the side effects of cancer drugs, researchers say.

In the animal study by researchers at the University Of Pittsburgh School Of Medicine And Magee-Womens Research Institute, previously frozen stem cells restored production of sperm that successfully fertilized eggs to produce early embryos.

Some cancer drugs work by destroying rapidly dividing cells. As it is not possible to discriminate between cancer cells and other rapidly dividing cells in the body, the precursor cells involved in making sperm can be inadvertently wiped out leaving the patient infertile, said senior investigator Kyle Orwig from Magee-Womens Research Institute.

"Men can bank sperm before they have cancer treatment if they hope to have biological children later in their lives," Orwig said.

"But that is not an option for young boys who haven't gone through puberty, can't provide a sperm sample, and are many years away from thinking about having babies," he said.

Even very young boys, though, have spermatogonial stem cells in their testicular tissue that are poised to begin producing sperm during puberty.

To see whether it was possible to restore fertility using these cells, Dr. Orwig and his team biopsied the testes of prepubertal and adult male macaque monkeys and cryopreserved the cells from the small samples. The monkeys were then treated with chemotherapy agents known to impair fertility.

A few months after chemotherapy treatment, the team re-introduced each monkey's own spermatogonial stem cells back in to his testes using an ultrasound-guided technique. Sperm production was established from transplanted cells in nine out of 12 adult animals and three out of five prepubertal animals after they reached maturity.

In another test, spermatogonial stem cells from other unrelated monkeys were transplanted into infertile animals, which created sperm with the DNA fingerprint of the donor to allow easy tracking of their origin.

In lab tests, sperm from transplant recipients successfully fertilized 81 eggs, leading to embryos that developed to the morula and blastocyst stages, which are the stages that normally precede implantion in the mother's uterus. Donor parentage was confirmed in seven of the embryos.

The 35 companies and researchers who spoke at last weeks stem cell conference in La Jolla faced a big challenge. They were given just 15 minutes each to pique the interest of potential investors and corporate partners.

To get that brief time in the spotlight at the Stem Cell Meeting on the Mesa, they came from across the country; two came from Europe.

Nearly all of them shared the goal of using stem cells and the bodys own regenerative ability to treat diseases and injuries. But their approaches to regenerative medicine vary greatly.

Many are tackling diseases such as diabetes, cardiovascular disease and hair regeneration. Some aim to provide new and useful stem cell types. Others provide equipment to expand regenerative medicines tool chest.

Here are five of the presentations:

Aderans Research Institute: Your own hair, back again. Thats the allure of Atlanta-based Aderans Ji Gami hair growth process. Patients donate hair-containing skin from their neck, and Aderans cultivates the hair follicle-forming cells. These cells are then injected back into the patient.

The cells stimulate new hair production in dormant hair follicles, working on male or female pattern baldness, said Aderans Chief Executive Ken Washenik. They have been tested in animals and are now being tested in people.

When we inject this mixture of dermal and epidermal cells we find that the cells intercalate (integrate) into existing hair follicle structures and reprogram them or turn them back on, Washenik said at his companys presentation.

Aderans has performed Phase 1 safety studies on Ji Gami, and has now started Phase 2 studies, looking for effectiveness.

DiscGenics: About 30 million Americans experience lower back pain, much of it caused by spinal disc degeneration. The market for treating it is about $4.4 billion, said Flagg Flanagan, president and CEO of DiscGenics, based in Salt Lake City.

Stem cell treatment may restore male fertility

Scientists say it may be possible to restore sperm production in boys who are left infertile by childhood cancer treatments, BBC News reported.

One of the harmful side effects of chemotherapy and radiotherapy is that the treatments also kill cells which make sperm. However, in a new study on monkeys, researchers from the University of Pittsburgh and the Magee-Womens Research Institute extracted sperm-producing stem cells before cancer treatment and later restored the cells.

Nine out of 12 adult monkeys and three out of five prepubescent monkeys were later able to produce healthy sperm again meaning the sperm was capable of fertilizing female eggs.

While male cancer patients have the option of freezing sperm before treatment, this is not an option for patients who have not yet goon through puberty.

"This report is a very useful step forward and clearly shows that the science of spermatogonial stem cells transplantation might one day work for humans, Dr. Allan Pacey, senior lecturer in andrology at the University of Sheffield, told BBC News. And, although the authors report relatively low efficiency so far, in the context of someone who does not have any banked sperm to fall back on, these odds are probably very encouraging to make this kind of approach worthwhile."

Pacey added a main concern of the treatment was that cancer could be lurking in the stem cells, raising the possibility of re-implanting cancer cells into a healthy patient.
