Avalon GloboCare Provides Updates on its Lead Scientific and Clinical Programs in CAR T-Cell Therapy and COVID-19 – GlobeNewswire

FREEHOLD, N.J., July 13, 2020 (GLOBE NEWSWIRE) -- Avalon GloboCare Corp. (NASDAQ: AVCO), a clinical-stage global developer of cell-based technologies and therapeutics, today announced an update on its four scientific research and clinical development programs in cell therapy and COVID-19 related initiatives, including AVA-001 and AVA-011 (FLASH-CAR), that leverage individualized CAR (Chimeric Antigen Receptor) T-cell therapy for immuno-oncology, as well as AVA-Trapfor mitigating COVID-19 cytokine storm, and S-layer based COVID-19 vaccine development.

AVA-001

Avalon has successfully completed a Phase I first-in-human clinical study of its leading CAR T-cell therapy candidate, AVA-001 (National Institute of Health clinical trial registration number: NCT03952923). AVA-001 is a third generation CAR T-cell therapy which involves the 4-1BB (or CD28) co-stimulation signaling pathway. Nine out of ten patients with relapsed/refractory B-cell acute lymphoblastic leukemia (B-ALL) have achieved complete remission (CR rate of 90%) within one month after receiving one dose of AVA-001 CAR T-cell therapy. The treatment with AVA-001 was generally well tolerated with minimal adverse side effects: no neurotoxicity or greater than Grade-1 cytokine release syndrome was observed in this cohort of patients treated with AVA-001. All patients who achieved CR successfully proceeded to allogeneic bone marrow transplant with curative intent. Accessory laboratory testing that accompanied this pilot clinical study has demonstrated evidence of enhancement in CAR T cell persistence and protection against CAR T cell exhaustion. This paradigm of bridging CAR T-cell therapy to bone marrow transplant creates a new therapeutic horizon with curative potential for patients with relapsed/refractory B-ALL and other hematologic cancers. Avalon is in the process of extending AVA-001 CAR T-cell therapy to the next phases of clinical development.

AVA-011 (FLASH-CAR)

Avalons FLASH-CARuses next generation CAR technology to modify patients T-cells and natural killer (NK) cells using a ribonucleic acid (RNA)-based platform rather than a viral vector.Avalons RNA-based CAR technology is designed to rapidly create personalized CAR therapies in 1 to 2 days compared to the 10- to 14-day bio-manufacturing time necessary to generate currently available CAR-T cellular immunotherapy. Avalons FLASH-CARtechnology is also designed to reprogram the immune cells to hone in on multiple crucial cancer cell targets, called tumor antigens, to potentially achieve superior therapeutic effect. Avoiding the use of viral vectors and complicated bio-processing procedures significantly reduces manufacturing costs, resulting in a more affordable and potentially breakthrough therapy for cancer patients. The FLASH-CARtechnology can also be used to generate off-the-shelf, universal cell therapy that has the potential to reach even more patients. Avalons first FLASH-CARplatform candidate, AVA-011, targets both CD19 and CD22 tumor antigens on cancer cells. Pre-clinical research on AVA-011, including tumor cytotoxicity studies, has been successfully completed and Avalon is immediately entering the process development stage to generate clinical-grade CAR-T and CAR-NK cells for use in human clinical trials. Avalon expects to begin a first-in-human clinical trial with AVA-011 for the treatment of relapsed/refractory B-cell lymphoblastic leukemia (B-ALL) and non-Hodgkin lymphoma in the first quarter of 2021. The goal is to apply AVA-011 as a bridge to bone marrow stem cell transplant therapy with curative potential for patients with these blood cancers.

Avalon GloboCare, has established the Avalon Combat COVID-19 Taskforce (ACCT) to expand and accelerate scientific/clinical development on multiple fronts to help combat the COVID-19 global pandemic through a strategic combination of therapeutic and vaccine approaches as follows:

AVA-Trap

Avalon has engaged with Massachusetts Institute of Technology (MIT) to co-develop a QTY therapeutic platform against the cytokine storm which causes lung damage and other organ failure associated with COVID-19. Cytokines are small protein molecules in the body required to regulate and maintain proper physiological functions. In some life-threatening diseases, however, cytokines are released in vast excess (also known as cytokine storm) leading to devastating damage to vital tissues and organs. A prime example is the widely recognized SARS-CoV-2 (COVID-19)-induced cytokine storm, which can lead to acute respiratory distress syndrome, lung fibrosis, multi-organ failure and death. The pre-clinical result has been published in the journal QRB Discovery (Cambridge University Press). Three USPTO provisional patents have been filed based on the unique QTY code protein design platform that uses six water-soluble variant cytokine receptors which have been successfully designed and tested to show binding affinity to the respective cytokines. Avalons AVA-Trap therapeutic program is currently entering animal model testing followed by expedited clinical studies with the goal of providing an effective therapeutic option to combat COVID-19 and other life-threatening conditions involving cytokine storms.

Intranasal and Oral COVID-19 Candidate Vaccine

Formed a strategic partnership program between Avalon GloboCare and the University of Natural Resources and Life Sciences (BOKU) in Vienna, Austria. The goal is to co-develop an S-layer vaccine that can be administered by an intranasal or oral route against SARS-CoV-2 (COVID-19). This partnership is led by BOKUs Professor Uwe B. Sleytr, an eminent member of the Austrian Academy of Sciences, who is a pioneer of applied surface layer (S-layer) nanotechnology, based on the repetitive protein structures that make up the outer surface of microbial cells. This vaccine strategy has the dual advantages of ease of manufacturing and delivery. The candidate vaccine is derived from a fusion of an S-layer viral particle mimic with the SARS-CoV-2 spike protein and could be delivered non-invasively via the nasal or oral passageways, rather than a needle-based injection into the muscle or under the skin. The S-layer protein-based vaccine is expected to both decrease the severity of a SARS-CoV-2 infectionpreventing the more severe respiratory inflammation and organ damage seen in many COVID-19 patientsand build immunity against the virus. Substantial progress has been made by developing the proprietary techniques necessary to synthesize conjugate vaccines consisting of an S-layer artificial viral envelope linked to a viral antigen, eliciting immune-protective antibody responses. Avalon plans to complete the proof-of-concept pre-clinical studies in 2020, followed by first-in-human clinical study of this candidate vaccine during 2021. In addition, Avalon plans to utilize S-layer technology to accelerate additional vaccine programs for other respiratory infections including different strains of the flu (influenza A/B), respiratory syncytial virus (RSV), and other viruses. Avalon is also actively exploring other practical uses of S-layer technology including targeted drug delivery, diagnostic devices, and therapeutic applications.

SMD – Scientists discover protective Alzheimer’s gene and develop rapid drug-testing platform – QMUL

The main challenge in testing Alzheimers drugs in clinical trials is that participants need to have symptoms. But once people have symptoms, it is usually too late for treatments to have a significant effect, as many brain cells have already died.

The only current way to test potential preventative treatments is by identifying participants who are at higher risk of developing Alzheimers and seeing if treatments prevent the onset of their disease. This includes people with Down's syndrome (DS) who have around a 70 per cent chance of developing Alzheimers during their lifetime. This is because the extra chromosome 21 they carry includes the gene for amyloid precursor protein which causes early Alzheimers when overdosed or mutated.

In the study, published in the Nature group journal Molecular Psychiatry, the researchers collected hair cells from people with DS and reprogrammed them to become stem cells, which were then directed to turn into brain cells in a dish.

In these brain-like cells, the researchers saw Alzheimers-like pathology develop rapidly, including the hallmark trio of signs of Alzheimers progression - amyloid plaque-like lesions, progressive neuronal death and abnormal accumulations of a protein called tau inside neurons.

Lead researcher Professor Dean Nietifrom Queen Mary's Blizard Institute commented: This work represents a remarkable achievement, as this is the first cell-based system that has the full trio of Alzheimers-pathologies, without any artificial gene overexpression. This system opens up the prospect for screening for new drugs aimed at delaying or even preventing Alzheimers before neuronal death starts.

The researchers showed that the system could be used as an early preventative-drug testing platform. They took two different drugs which are known to inhibit -amyloid production, tested them on these brain cells, and in six weeks showed that they prevented the onset of Alzheimers-pathology.

Although these two particular drugs have failed clinical trials for other reasons and therefore arent suitable treatments for Alzheimers, the team showed the proof-of-principle that the system can be used on any drug compound, and within six weeks show whether or not it has potential for further investigation.

The team also found proof of the existence of a naturally-functioning Alzheimers suppressor gene (BACE2 gene). Acting in a similar way to tumour suppressor genes in cancer, the increased activity of this gene contributes to the prevention/slowing down of Alzheimers in human brain tissue, and could in the future be used as a biomarker to determine peoples risk of developing the disease, or as a new therapeutic approach by boosting its action.

Professor Dean Nietiexplained: Although its still early days, the system raises a theoretical possibility for further development as a tool to predict who might develop Alzheimers. The same stem cell process could be used on anyones hair follicles, the resulting brain cells of which may or may not then develop Alzheimers-pathology in the dish. The idea would be to catch the people at higher risk of early disease in a cell-based system, before it starts in a persons brain, and allow for the possibilities of individualised preventive interventions. We are still a long way from reaching this goal.

Co-author Professor John Hardy from UCL added: I think we have the potential now to develop a new, human model of the disease which would be a great step forward.

The discoveries in this study were dependent on contributions from people with DS who kindly accepted to participate in this study, the results of which could be beneficial for people with and without DS in preventing Alzheimers. The Downs Syndrome Association (UK) provided essential support and help with recruitment of the participants in the study.

The Downs Syndrome Association is delighted to have been able to support this brilliant work.

The main funders of the project were Wellcome (UK) and National Medical Research Council (Singapore), and involved major contributions from the Nanyang Technological University Singapore, UCL (Prof. John Hardy and Prof. Henrik Zetterberg) and Sahlgrenska Academy of the University of Gothenburg, Sweden (Prof. Henrik Zetterberg), as well as a number of international partners, including The Croatian Institute for Brain Research at the University of Zagreb Medical School.

Michael Longaker won a national title at Michigan State, then dove into the world of medicine – MLive.com

Four decades later, the memory of his first practice against Earvin Magic Johnson is a vivid one for Michael Longaker.

As a sophomore guard for Michigan State in 1977-78, Longaker was among those tasked with defending the Spartans star newcomer. He spent that day exhausting himself while chasing the bigger and more athletic Johnson around Jenison Fieldhouse.

But the challenges didnt stop even when practice did. After leaving the court, Longaker balled up the tape on his ankles and threw it in a wastebasket. Johnson promptly grabbed the ball of tape and boasted he could sink the wastebasket shot from further away.

I was like Oh my gosh, what a nightmare, Longaker said. That was a window into how competitive Earvin was at everything.

While they competed in practice, the two teamed up to help Michigan State win the schools first national title in 1979. Johnson was the face of the team, while Longaker was a role player helping keep Johnson and others sharp in practice every day.

After Michigan State, the two teammates went to California and found plenty of professional success.

Johnson, of course, won five NBA titles and three MVP awards in his Hall of Fame career for the Lakers.

Longaker, meanwhile, settled in Northern California and became a leader in a different field: medicine.

I do something very different from my teammates, I imagine, Longaker said this week from his office.

Longaker has spent the last two decades at Stanford University, where hes a professor at the School of Medicine and the co-director of the Institute for Stem Cell Biology and Regenerative Medicine. His work focuses on scarring and how to regenerate skin and improve wound healing.

Its a field of study far different than the matchup zone he worked to master under Jud Heathcote. But playing and studying at Michigan State, he said, set him up for a successful career in medicine.

I cant overstate the role that Michigan State has played in my career, Longaker said.

Longaker spent four years at Michigan State working hard in practices like that first one and fighting for playing time on some immensely talented Spartans teams.

His contributions were often not seen on box scores. Later in his career, Heathcote started asking Longer for his thoughts before and during games. He saw Longaker as a potential future coach.

Longaker, though, had other career plans. He initially planned on going into dentistry, but a summer spent doing research with James Potchen, then the chair of Michigan States Department of Radiology, sold him on a career in medicine.

Heathcote ended up writing a letter of recommendation that helped Longaker get into Harvard Medical School. But even then, he still held out hope to see his former player on the sideline one day.

I said Coach, Im kind of committed to being a physician, Longaker said.

Longaker finished medical school with the intent of becoming a pediatric heart surgeon. But during a year of research during his residency at the University of California, San Francisco, he was assigned to work under a doctor who operated on children before they were born and asked to investigate how embryos heal wounds.

His findings, that embryos heal without scars in their first two trimesters, set his career on a different course. One year of research turned into four, then was followed by more training in New York and Los Angeles before he landed at Stanford in 2000.

Now, most of his work is in directing a lab that focuses on skin and the skeleton, developing techniques to use stem cells to improve wound healing and prevent scarring. His work has earned him numerous awards and hes been an inventor on over 40 patents and patent applications.

Its been great, Longaker said. I couldnt have predicted it.

That world is far removed from that of Michigan State basketball, but Longaker finds ways to bridge his past and his present. He has two teenage sons who follow the program closely he was elated this week in the afterglow of commitments from Emoni Bates and Max Christie and typically attends multiple games per year. Tom Izzo didnt join Heathcotes staff until after Longaker left, but the two have developed a friendship over the years.

Longaker has also become involved with the sports program at his employer (he attended the 2013 Rose Bowl wearing a split T-shirt featuring both Stanford and Michigan State) Former Stanford running back Bryce Love, the 2017 Heisman Trophy runner-up, worked in Longakers lab while completing his degree in human biology, and Oscar da Silva, an All-Pac 12 forward for the Cardinal, counts Longaker as his academic mentor.

One of Longakers recent trips back to East Lansing was last year, for the 40th anniversary of the 1979 national title. And when that team meets again in another decade, Longaker hopes his smarts can help reverse the outcome of that first practice way back when.

I always joked with my teammates, at our 50-year reunion, Im going to dominate, Longaker said.
