Nilanjan Ghosh, MD, a hematologist and medical oncologist at the Levine Cancer Institute, Atrium Health, discusses the effect coronavirus disease 2019 (COVID-19) had on using chimeric antigen receptor (CAR) T-cell therapy to treat patients with diffuse large B-cell lymphoma (DLBCL).

Ghosh says that when the pandemic first started, he and other physicians thought it would not only affect CAR T-cell therapy but also stem cell transplants. CAR T-cell treatments for DLBCL include therapies such as axicabtagene ciloleucel (Yescarta) and tisagenlecleucel (Kymriah). There was a lot of uncertainty around how long the pandemic would last since it started abruptly with many acute cases, which filled intensive care units and hospital beds.

As the months went on, physicians adapted to treating patients during COVID-19, which Ghosh calls the new normal. As of October, the pandemic was not affecting the delivery of CAR T-cell therapy for clinical trials and treatment with standard of care in practices. At the beginning of COVID-19, he and his colleagues were doing their best to deliver standard-of-care CAR T therapy, but it did affect CAR T-cell delivery for clinical trials because of regulatory constraints. There were several months where accrual for these trials was limited, but over the past months, many of those constraints have been lifted and the clinical trials have opened back up.

In connection with the reorganization, Lance Alstodt has been appointed the Companys President, Chief Executive Officer and Chairman of the Board. Mr. Alstodt said, This process has been a long and challenging journey for the Company. Im inspired by the great resolve and execution from our employees, professionals and investors. We are very pleased that all requirements have been met for us to emerge. Allowed creditor claims have been fully satisfied and, as importantly, our equity holders have retained their shares in this exciting new opportunity. We were able to preserve all of our intellectual property assets and look forward to initiating our Phase 2 clinical trial.

Based upon the Companys emergence from Chapter 11 reorganization, FINRA has removed the Q at the end of its trading symbol. Shareholders do not need to exchange their shares for new shares.

Initial preclinical research indicates that increased amounts of brown fat in the body may be responsible for additional caloric burning as well as reduced glucose and lipid levels. Researchers have found that people with higher levels of brown fat may have a reduced risk for obesity and diabetes.

ABSTRACT

INTRODUCTION: The purpose of this study was to determine if pharmacological treatment could increase progenitor cell proliferation in the Sub-ventricular Zone of aged rats. Previous work had shown that increasing progenitor cell proliferation in this region correlated well (R2=0.78; p= 0.0007) with functional recovery in a damaged corpus callosum (white matter tract), suggesting that progenitor cell proliferation results in oligodendrocytes in this region.

METHODS: 10 month old male and female Sprague Dawley rats were fed the drugs for 30 days in cookie dough, then immunocytochemistry was performed on coronal brain sections, using Ki67 labeling to determine progenitor cell proliferation.

RESULTS: Female rats showed low endogenous (control) progenitor cell proliferation, significantly different from male rats (P<0.0001), at this age. Ascorbic Acid (20 mg/kg, daily for 30 days) increased progenitor cell proliferation overall, but maintained the innate gender difference in stem cell proliferation (P=0.001). Prozac (5 mg/kg, daily for 30 days) increased progenitor cell proliferation for females but decreased stem cell proliferation for males, again showing a gender difference (P<0.0001). Simvastatin (1 mg/kg for 30 days) also increased progenitor cell proliferation in females and decreased progenitor cell proliferation in males, leading to a significant gender difference.

DISCUSSION: The three drug combination (fluoxetine, simvastatin, and ascorbic acid, patent # 9,254,281) led to ~ 4 fold increase in progenitor cell proliferation in females, while male progenitor cell proliferation was highest with 50 mg/kg ascorbic acid. However, the ascorbic acid increase in proliferation appears to be only on the sides of the ventricles, which is not the region that normally gives rise to oligodendrocytes.

CONCLUSION: There are innate gender differences in progenitor cell proliferation at the Sub-Ventricular Zone at middle age in rats, possibly due to loss of estrogen in females. We also see notable gender differences in progenitor cell proliferation in the Sub ventricular Zone in the response to common drugs, such as fluoxetine, simvastatin and Vitamin C (ascorbic acid).

OSS, Netherlands, Nov. 20, 2020 /PRNewswire/ -- Glycostem Therapeutics B.V., a leading clinical-stage company focused on the development of therapeutic off-the-shelf Natural Killer (NK) cells, today announced the opening of a phase I/IIa trial for its lead product oNKord for the treatment of AML patients. The WiNK trial will enroll 33 AML patients at eight clinical sites based in five European countries. oNKord is the company's first-generation off-the-shelf allogeneic NK cellular immunotherapy product. Glycostem is furthermore developing a range of second (CAR-NK) and third generation (TCR-NK) NK products in-house.

"During our phase Isafety study, in which ten elderly AML patients were treated, we verified our lead productoNKordto be a safe therapeutic agent in the field of cellular immunotherapy. Interestingly, year 1 survival data showed promising efficacy outcome (80%) compared to historical cohort data (40%)," tells Troels Jordansen, CEO at Glycostem. "The opening of this phase I/IIa / pivotal trial is an important step and we are looking forward to the data from this study, which will allow us to draw conclusions regarding oNKord safety and tolerability and most importantly its efficacy for treatment of AML patients."

Professor Doctor Arnold Ganser, MD of Hannover Medical School in Germany and WiNK trial coordinating investigator said: "The preliminary efficacy data obtained with oNKord was very promising and NK-cells have proven to be highly capable of targeting cancer cells. Therefore, I am excited about participating and being able to enroll patients in the WiNK trial." He continues: "Currently there is a risk of relapse for 80% of the successfully treated patients. This new form of cellular treatment can change the lives and future prospects of many patients with AML worldwide who do not proceed to allogeneic hematopoietic stem cell transplantation, in particular elderly patients, who represent the majority of patients with AML."

WiNK is a prospective two-stage, open-label, single arm, multicenter phase I/IIa trial to evaluate the safety and efficacy of oNKord, an off-the-shelf, ex. vivo-cultured allogeneic NK cell preparation, in 33 adults with AML who are in complete morphologic remission with residual measurable disease and with no strong indication for hematopoietic stem cell transplantation.
