Collaboration to develop stem cell therapies for COVID-19 and future pandemic diseases

Avacta Group plc , the developer of Affimer biotherapeutics and reagents, has announced an expansion of its collaboration and license agreement with Daewoong Pharmaceutical Co. Ltd. and AffyXell Therapeutics, the joint venture established in South Korea by the two companies, to develop stem cell treatments incorporating Avactas neutralizing Affimer therapy for the treatment of seriously ill patients with COVID-19 and to also prepare to rapidly develop similar therapies for future global pandemics.

Respiratory diseases such as COVID-19 can cause serious damage to the lungs as a consequence of over-activation of the patient's immune system, resulting in cytokine release syndrome that can potentially lead to multiple organ failure and death. Stem cell therapies offer a very promising approach to repair the damage to lung tissues in these pulmonary diseases by controlling the immune balance.

AffyXell Therapeutics, the next-generation cell and gene therapy joint venture between Avacta and Daewoong Pharmaceutical, is developing a novel class of mesenchymal stem cell (MSC) treatments that are engineered to also produce Affimer therapies in the patient at the site of action.

The expansion of the agreement between Avacta, Daewoong Pharmaceutical and AffyXell announced today extends the scope of the partnership to include Affimer molecules that target viruses, such as coronaviruses, in order to develop therapies that repair the lung damage caused by COVID-19 whilst also producing neutralizing Affimer molecules to prevent the progression of the disease.

AffyXell will engineer mesenchymal stem cells to express SARS-COV-2 neutralizing Affimer molecules in order to develop treatments for seriously ill COVID-19 patients, and will also prepare for rapid development of next-generation stem cell therapies for future infectious respiratory disease outbreaks.

Mesoblast sees lasting need for COVID treatment beyond a vaccine – Sydney Morning Herald

Mesoblast is hoping its flagship product remestemcel-L will be shown to relieve acute respiratory distress syndrome (ARDS) in COVID-19 patients. The company is running a phase 3 trial of the product in US hospitals, but the research is still in its early stages, with a pilot study of the project looking at only 12 patients.

The US Data Safety Monitoring board, which reviews clinical data, had selected a date in early September to complete an initial review of Mesoblast's phase 3 trial, the company said. There are no approved treatments for this kind of respiratory distress, and it is not yet known whether its product will prove safe and effective in a large group of patients.

Dr Itescu said the company would know the results by the end of this calendar year. If the project is successful, the product could be manufactured in Australia for export into global markets to treat COVID patients around the world.

"We're tracking pretty much as expected," Dr Itescu said.

The company has also been working on getting the same product approved in the US for use in children who have graft-vs-host disease, a complication from bone marrow transplants.

The Food and Drug Administration will review data about the effectiveness of the treatment on August 13. Dr Itescu said the regulator would advise whether the product has received approval before September 30.

Reviews of these two projects could have large implications for Mesoblast's future.

The company, which has been backed by high-profile investors including billionaire Alex Waislitz, has seen its shares turbocharged by updates on its COVID research in recent months. They jumped more than 5 per cent again on Thursday, trading at $3.84 by mid-afternoon.

The business generated $US2.1 million from customers in the June quarter and had $19.6 million in operating expenses.

UAE leads Covid-19 research in the Middle East – The National

Since the onset of the coronavirus pandemic, the UAEs efforts to combat its spread and prepare for a post-lockdown reality have made it a positive example for the region and the world.

Public health officials have carried out nearly 5 million coronavirus tests over the past six months a remarkable achievement in a country with a population of 9 million. This mass-testing strategy has curbed the spread of the virus and protected the population.

Another pillar of the Emirates success thus far is the willingness to invest in local expertise and drive resources towards Covid-19 research. In May, QuantLase Imaging Lab, which is part of International Holdings Company in Abu Dhabi, announced it had developed a rapid-testing technology capable of diagnosing coronavirus in mere minutes.

The Covid-19 screenings use a diffractive phase interferometry (DPI) test, which can detect early signs of the body fighting a virus. A centre has been set up on the border between Dubai and Abu Dhabi to facilitate travel into the latter, where a negative coronavirus test remains a requirement for entry.

The facility carries out thousands of tests a day, with more DPI screening centres set to open soon.

Abu Dhabi and Al Ain are also the first cities in the world to commence Phase III trials for a World Health Organisation-enlisted coronavirus vaccine. More than 10,000 people have volunteered for the final-stage trials, which aim to identify antibodies that provide immunity against the virus. Only three out of 20 coronavirus vaccines are currently in Phase III trials globally.

While the world awaits a successful vaccine, the Emirates is developing new treatments locally. In May, Abu Dhabi Stem Cell Centre announced it had produced a stem cell therapy treatment that helps to alleviate Covid-19 symptoms. The centre has thus far treated 2,000 patients free of charge.

This week, a one-month study of coronavirus immunity was launched in Abu Dhabi. The tests used will detect whether a person has been infected with coronavirus in the past, providing scientists with data on how widespread infections have been, as well as the prevalence of asymptomatic carriers. Such results will be instrumental in designing future public health policies.

The ground-breaking developments have been made easier by the fact that the UAE was already heavily focused on becoming a leading centre for medical research in general. Now, it is facilitating advancement in other areas of medicine, demonstrating the synergies that can form when developing a global medical hub.

This week, Abdullah Nazir Ahmad Muhammad became the first patient in the UAE to undergo bone-marrow transplant, in Sheikh Khalifa Medical City, Abu Dhabi. The transplant used stem cell technology originally developed to treat Covid-19 infections, and as a result the blood-cancer patient was given a second chance of life.

The UAEs experience thus far provides an important lesson for public policymaking: that tackling challenges as daunting as a viral pandemic takes long-term planning and investment in strategic sectors. The same qualities are required in order to deal with a pandemics aftermath.

Ghadan 21, a set of large-scale reforms and projects launched last year by Sheikh Mohamed bin Zayed, Crown Prince of Abu Dhabi and Deputy Supreme Commander of the UAE Armed Forces, has helped to diversify the economy and develop non-oil sectors. Early investment in education to foster local talent has long attracted the brightest minds from around the world. When the pandemic struck, the country already had a solid healthcare infrastructure.

Next year marks the 50th anniversary of the UAEs founding. Seen through the larger lens of human history, five decades is a very short period of time. But as the UAEs pandemic experience shows, no matter how far back a nations history stretches, it must make use of the time it has to prepare for a brighter future.
